The story of Kisi Ka Bhai Kisi Ki Jaan revolves around Bhaijaan, a courageous man who loves to live with his three brothers, Ishq, Moh, and Love. He is against getting married, as he thinks it will not be good for his relationship with his brothers. The three brothers of Bhaijaan also pretend to be against marriage. Still, all three are managing their secret relationships with Sukoon, Muskaan, and Chahat.

Now, to get married to these girls, the brothers find it necessary to make Bhaijaan fall for a girl. Here comes Nadeem Chacha, who told the three brothers that Bhaijaan used to love a girl named Bhagyalakshmi in the past. However, when Bhaijaan's brothers conspire to reunite Bhaijaan with Bhagyalakshmi, they find out she is already married and has a son.

Then the brothers try to make Bhaijaan fall for another girl named Bhagyalakshmi, as Bhaijaan is more in love with the name "Bhagyalakshmi.'' The idea worked, and both fell in love.
